Antibiotic resistance of pathogenic bacteria is a rising threat to human health world-wide. For treatment and prevention it is crucial to identify and track antibiotic resistant bacteria. X-Shining AST Ampicillin (AST stands for antibiotic susceptibility test) is an alternative test system to conventional, growth based AST tests with a read-out time of only 30 to 60 min. The test relies on the monitoring of released ATP in the absence and presence of the Ampicillin at 10 µg/mL final assay concentration. ATP release is measured via luminescence signal generated by reaction of ATP with X-Shining luciferase and D-luciferin. Product EX182071 is a lyophilisate containing all necessary components required for the assay and can be used immediately after reconstitution with ultrapure water. The antibiotic Ampicillin interferes with cell wall synthesis, which leads to increased leakage of ATP within 30 to 60 min after exposing bacterial cells to nutrients and antibiotic. Thus, Ampicillin sensitive bacteria exhibit a considerably increased luminescence (at least factor 10) in the presence of Ampicillin relative to control assays without antibiotic, using product EX182068, X-ShiningTM Base Medium. By contrast, resistant strains which express carbapenemase do not show such a response, the ratio of luminescence with and without antibiotic remains below 3.
