Retaspimycin hydrochloride (RTH) is a protein-based inhibitor of tumor cell growth. It binds to the response element on the 17-aag gene and inhibits the synthesis of this protein. RTH also inhibits alcohol dehydrogenase, which may contribute to its antitumor activity. Retaspimycin hydrochloride has been shown to inhibit squamous cell carcinoma in humans and other cancers in mice. The mechanism of action may be due to its ability to bind to folate receptor and reduce the expression of HSP90, an important protein for cancer cell survival.
